下调NDRG1 表达对胰腺癌细胞增殖与凋亡的影响

摘要

目的:探讨NDRG1 在胰腺癌细胞中的表达及其与MMP-7 的关系。 方法: 检测NDRG1 与MMP-7 蛋白与mRNA 在4 种胰腺癌细胞株(PANC-1、BXPC-3、CAPAN-2、 SW1990)表达;构建靶向NDRG1 的干扰质粒siNDRG1,转染PANC-1 细胞,检测转染后PANC-1 细 胞NDRG1 与MMP-7 蛋白与mRNA 的表达,以及增殖与调亡情况。 结果:4 种细胞株中均有NDRG1 与MMP-7 蛋白与mRNA 的表达,且两者的表达水平随着细胞分化程 度的降低而升高(均P<0.05);PANC-1 细胞转染siNDRG1 后,NDRG1 与MMP-7 的蛋白与mRNA 表 达均明显降低、细胞增殖明显抑制、凋亡率明显升高(均P<0.05)。 结论:NDRG1 的表达与胰腺癌细胞的分化程度密切相关,且可能通过上调MMP-7 表达促进胰腺癌细 胞的生长。

Influence of down-regulation of NDRG1 expression on proliferation and apoptosis in pancreatic cancer cells

Abstract

Objective: To investigate NDRG1 expression and its relation with MMP-7 in pancreatic cancer cells. Methods: The protein and mRNA expressions of NDRG1 and MMP-7 in 4 different pancreatic cancer cell lines (PANC-1, BXPC-3, CAPAN-2 and SW1990) were determined. The interference plasmids siNDRG1 were transfected into PANC-1 cells after their construction, and then the protein and mRNA expressions of NDRG1 and MMP-7, cell proliferation and apoptosis after transfection were determined. Results: All the 4 cell lines presented either the protein or mRNA expressions of both NDRG1 and MMP-7, and their expression levels were increased with decrease of the degree of cell differentiation (all P<0.05). In PANC- 1 after siNDRG1 trasfection, the protein and mRNA expressions of both NDRG1 and MMP-7 were reduced, proliferation was suppressed, and apoptosis rate was increased significantly (all P<0.05). Conclusion: NDRG1 expression is closely related to the degree of differentiation of pancreatic cancer cells, and may probably promote cell growth through up-regulating MMP-7 expression.
